Non-invasive Liquid Biopsy

A non-invasive liquid biopsy permits the analysis of multiple circulating biomarkers in one sample and facilitates the discovery of disease as well as treatment response monitoring in applications such as oncology and prenatal analysis.
We have developed Dynabeads® magnetic beads for enrichment of circulating tumor cells (CTCs), both for positive and negative selection. For a positive selection Dynabeads® magnetic beads pre-coupled with EpCAM (epithelial cell adhesion molecule) were used to capture circulating epithelial cells. For negative selection, a combination of Dynabeads® CD45 and Dynabeads® CD15 gave a 95 - 99 % depletion of mononuclear cells.
In addition, we have developed Dynabeads® magnetic beads for capture of exosome sub-populations from pre-enriched cell culture supernatant, by coupling Dynabeads® with antibodies targeting CD81, as well as CD9 and CD63 (data not shown).
Finally, we aim to provide a universal solution for high-throughput automation for efficient isolation of circulating cell-free DNA (cfDNA) isolation from large volumes of serum or plasma by using Dynabeads® MyOneTM SILANE. The bead surface chemistry in combination with the Proteinase K-free workflow enables highly efficiency capture of DNA in the <220 base pair range compared to other methods, providing the added advantage of a quick, reproducible, and cheap enrichment of circulating cfDNA.
