Insightful Advice for Avoiding Clonal Deletions

The latest IDT DECODED newsletter provides recommendations for successful DNA cloning.

Integrated DNA Technologies (IDT) offers valuable advice in the October edition of DECODED to help avoid deletions that sometimes occur during cloning.

For reasons that are not well defined, scientists who use oligonucleotide primers for PCR amplification prior to cloning occasionally obtain sequence deletions across multiple clones, often affecting the same, singular base.

While site-directed mutagenesis can resolve cloning deletions (see the free IDT Mutagenesis Application Guide for help with this approach), optimizing the process to avoid deletions is often more desirable, saving time and cost.

The cloning tips from IDT to increase the odds of generating correct clones are featured in the latest edition of the DECODED newsletter.
