Bioo Scientific has recently launched the NEXTflex™ Small RNA-Seq Kit v3, which improves small RNA analysis by making the library prep protocol completely gel-free or allowing for low-input small RNA library preparation.
This kit also reduces the biases associated with small RNA sequencing by using patent pending randomized adapter technology, resulting in small RNA libraries with more accurate sequencing data and a dramatic reduction in bias.
Gel purification steps in library prep are undesirable because they are slow, can result in sample loss, and reduce researchers’ ability to automate library prep. While magnetic beads have been incorporated to replace most gel steps in other types of library prep, most small RNA library prep protocols still required an acrylamide gel size selection for library purification.
The new NEXTflex Small RNA-Seq Kit v3 has incorporated a patent pending method which allows the gel size selection step to be replaced with a bead purification step with input of 200 ng -2 µg total RNA or purified small RNA from 200 ng-10 µg total RNA.
This same patent pending innovation facilitates small RNA library prep from input as low as 5 ng of total RNA. However, when library prep is preformed using less than 200 ng of input, a gel size selection step is typically required.
The NEXTflex Small RNA-Seq Library Prep Kit v3 incorporates randomized adapter technology to reduce bias introduced during the ligation step of library prep. The ligation step of small RNA library prep is ordinarily a source of severe bias.
The randomized adapter strategy incorporated into the NEXTflex Small RNA-Seq Kit v3 allows small RNAs of any sequence to “find” their respective optimal adapters, resulting in small RNA libraries with a dramatic reduction in bias.
