Example Application: PIPETMAX profiling mechanisms of toxicity
Accuracy and reproducibility are key to verifiable science. Automating the steps of the mechanistic toxicity workflow including cell plating, compound titration and detection reagent addition to 384-well microplates is done with PIPETMAX. Multiple assay plates can be assembled, allowing for the assessment of both time and dose-dependent effects. The detection reagents consist of four assays added simultaneously to the assay plate, each measuring various biomarkers associated with cell viability or toxicity.
Dead cell protease, caspase-3/7 and DNA staining are biomarkers associated with cytotoxicity and show an increase in signal over time. Dead cell protease and DNA staining correlate with changes in membrane integrity, while caspase-3/7 correlates with cells undergoing apoptosis.
