Nucleofection® of mouse ES cells

Efficient delivery of DNA is crucial for gene targeting in embryonic stem cells and for successful generation of knock-out mice. Data presented in this technote show that Nucleofection® leads to a homogenous gene expression pattern in mouse ES cell colonies. In contrast to standard electroporation, Nucleofection® results in a 100% deletion of frt-flanked genes on both alleles. Viable and fertile mice were generated from transfected mouse ES cell clones.
