The complete system allows the user for the first time to use temperature as another dimension for FFF separation. The separation in FFF is based simply on the diffusion of parts (Brownian molecular movement) and a temperature control brings up a large variety of advantages. The new AF2000 MT-MALS-Series offers such a temperature control and therefore reaches a new level of performance.

The AF2000-MT connected to a MALS-lightscattering detector is a completely new developed and optimized analytical system based on Asymmetric Flow Field-Flow Fractionation (AF4) and Multi-Angle Light Scattering (MALS). The separation range covers a very broad molar mass and particles size range. The separation itself is a very gentle procedure without any shear forces. The online connected and completely in the AF4 software integrated Multi-Angle Light Scattering-Detector makes it possible to measure the molar mass and the particles size directly without the usage of calibration standards.

The AF2000-MT-MALS Serie has a technically similar setup like a GPC-MALS-System but avoids the common limitations of chromatography. So the in liquid chromatography used column is in a AF2000 MT-MALS system replaced by a specially patented separation channel who has no stationary phase. The retention happens only based on size and by the influence of a cross flow as a separation force.
Proteins, Antibody, Aggregates, Conjugates, Viruses, Liposomes, Starches, Hyaluronic Acids, Alginates and many other Biopolymers but also a large variety of nanoparticles like Gold, Carbon-Nano-Materials and Latexparticles,
can be separated in a range of 10E3 - 10E9 Da and 1 nm - 10 µm . In contrast to Column chromatography the Mid Temperature Flow FFF has no size exclusion limit which would reduce the separation range for high and ultra high molar masses. Through the absence of any stationary phase and free choice of the solvent any unwanted adsorption effects can be avoided.

Unique Advantages
Ultrahigh resolution and very broad size range for polymers and particles
No shear forces and gentle separation especially for biomolecules
Direkte molar mass and particle size determination without calibration standards
Temperature controlled system with patented reusable separation channel
No stationary phase or columns as spare part necessary
Analytes can be directly fractionated, collected and measure with additional on- and offline analytical methodes as Fluorescence, EM, AFM, etc.

The Postnova AF2000-MT-MALS-System consists of the modular FFF separation system and the integrated into the software connected MALS-/UV- and RI-Detectors. The unique NovaFFF software platform controls the complete FFF-Detector system. But the software also collects the data and also the direct calculation of the molecular weight and the particle size. Additional detectors as DAD, Fluorescence and ELSD can be also connected.

The AF2000 MT-MALS Series is a basic tool now for any researcher and quality assurance manager, who has to solve questions in the area of Proteins, Biopolymers and Nanoparticles. The technology has the special advantage of a very broad separation range which can never be reached with classical chromatographic methods combined with a high separation technology for proteins, bio-/polymers and nanoparticles. This allows the separation of these analytes within one sample with a higher resolution and characterization. Additionally the common disadvantages of packed columns are avoided.
