Differentiation and characterization of subvisible particulates in therapeutic protein products

The formation of protein aggregates is a particular concern for parenteral administration biopharmaceuticals due to the potential for increased immunogenicity.

An Introduction to Proteins in 30 minutes

In the biotechnology industry, the majority of research is directed towards characterizing the structure and function of proteins. New methods are continually being developed to accomplish this.

Performing ADCC Reporter Bioassay with an Environmentally Controlled Microplate Reader

The Promega ADCC Reporter Bioassay provides effector and target cells in frozen, thaw-and-use format, removing the need for isolating or culturing cells in order to perform the assay. Reporter induction requires a six-hour to overnight incubation in a standard cell culture incubator. The CO2 and temperature control features of the Tecan Infinite® 200 PRO with GCM allow the assay induction to be performed without the requirement for a cell culture incubator.

Tag-lite® two-cell assay: a valuable tool for protein drug discovery

Antibodies are central mediators of the immune system, IgG being the most dominant. Binding of IgG/antigen complexes to membrane-bound Fc? receptors triggers a cellular immune response. Alterations in the Fc portion of the antibody by introducing mutations can lead to complete abolition of Fc? receptor binding. This describes the development of a cellbased TR-FRET assay to determine the simultaneous binding of an antibody to its antigen and huFc?RIIIa present on different cells.

3D Cell Culture: mimicking real tissue

Technological advances in the last couple of years have enabled 3D scaffolds to be engineered that offer a real proxy for the in vivo environment which can be easily and routinely used in a cell culture laboratory.

Alvetex Scaffold enables cells to develop natural, in vivo-like intercellular interactions, providing an ideal environment for real three-dimensional growth and nutrient exchange that is comparable to intra-capillary exchange in living tissues.

Protein Purification Workflow Development Using Bio-Rad's NCO' Chromatography System

High purity protein is a common requirement for biochemical and structural studies. A common approach is to recombinantly express an affinity-tagged version of the protein of interest. This is, however, not always a viable option. This application note discusses protein purification workflow development for untagged proteins and introduce a new indicator of method performance, the purity quotient difference.

Direct Visualisation, Sizing and Counting of Aggregation in Proteins

Characterising the state of aggregation in proteins is of paramount importance when trying to understand biopharmaceutical product stability and efficacy. Product quality, both in terms of biological activity and immunogenicity can be highly influenced by the state of protein aggregation.

Direct Visualisation, Sizing & Counting of Viruses and Phages in Liquid Suspension

A new laser-based nanoparticle tracking analysis system is now available which allows nanoscale particles such as viruses and virus aggregates to be directly and individually visualised in liquids in real-time, from which high-resolution particle size distribution profiles can be obtained.
Contaminant bovine IgG assay
Application Note

Contaminant bovine IgG assay

Fetal calf serum containing bovine IgG is commonly used in mammalian cell culture media for the production of recombinant pharmaceuticals. During the purification process of a recombinant pharmaceutical, bovine IgG may be copurified with the product, and may represent a biological hazard for clinical use. Therefore, it may be necessary to assay for contaminant bovine IgG in a final product originating from mammalian cells.

Dual-Glo Luciferase Reporter-Gene Assay on the SpectraMax M5, LMax II384 and Analyst GT Microplate Readers

Reporter-gene assays are used to study eukaryotic gene expression. Dual genetic reporters are commonly used in transient transfections of cultured cells to minimize experimental variability. Recently, Promega introduced a Dual-Glo Lucoferase assay system which replaced the outdated ‘flash’ assays used previously. Ensuring both genetic reporters have different substrate requirements enables them to be detected using a single reaction mixture.
