Intellicyt® T Cell Memory Cell and Cytokine Profiling Kit

T cells are generated and released from the thymus as naive T cells (TN). After infection or antigen encounter, the TN cells proliferate and differentiate into effector T cells (TE) which travel to site of infected target cells for destruction, hence clearing out the infection. After the infection is cleared, the majority of the TE die, but a small percentage develop into long lived resting state memory T cells (TM). These TM cells accumulate throughout life and have the capability to provide life-long protection against a number of pathogens. The positive or negative expression of certain surface markers such as CD45RA, CD45RO, CD27, CD62L and CD95 identify multiple T cell subsets at different stages of differentiation after activation. In addition, functional analysis of the cells are assessed by quantitating the levels of secreted IFNγ and IL-10. Understanding the biology of T cell memory could be useful for adoptive T cell therapy workflows and will enable scientists to design more effective vaccines for human diseases.

Kit Description

The Human T Cell Memory Cell and Cytokine Profiling Kit measures T cell memory phenotype and function at different stages while providing information about their health and their role in cytokine secretion. This one wash assay requires minimal hands on time and measures both cells and beads to provide the following biological insights:

  • Live immune cells are distinguished from dead cells by staining with a fluorescent membrane integrity dye.
  • Cells are immunophenotyped by staining with a fluorescent antibody panel to separate CD3+ T cells, CD3- non-T cells, CD4+ T helper cells, CD8+ T cytotoxic cells.
  • The panel also includes 5 different T cell surface markers for T naive/memory/effector cell phenotyping:  CD45RA, CD45RO, CD27 (long term survival), CD62L, and CD95.
  • Optimized workflow enables the measurement of effector cytokines secreted by activated T cells, including pro-inflammatory cytokine IFNγ and anti-inflammatory cytokine IL-10 in the same assay well.
