The requirement for better in-vitro models that are compatible with high-throughput screening campaigns has led to the development of 3D cell cultures models, especially muliticellular spheroids, which retain many of the morphological and genetic traits of tumours.
Here we describe the formation of such spheroids by two methods: on ultra-low attachment plates and in semi-solid agarose. Both methods are compatible with 96- and 384-well microplate formats.

Acute Infective Endocarditis: Early Surgery versus Conventional Treatment
Infective endocarditis (IE) refers to thew inflammation of the inner layer of the heart, the endocardium, secondary to an infectious (most commonly bacterial) agent. It affects the left side of the heart more commonly than the right. IE has an estimated incidence of 1.4 to 6.2 per 10,000 persons in the developed countries, and is most commonly caused by Staphylococcus aureus, coaguase-negative staphylococcus, Viridans group streptococci and Straptococcus bovis.

Automation of a Generic Fluorescence Methyltransferase Activity Assay
Epigenetic processes are attracting considerable attention in drug discovery as their fundamental roles in controlling normal cell development and contributions to disease states become more clearly defined. This work combines a fluorescence-based assay with liquid handling and dispensing instrumentation and a multi-mode reader which can be used to monitor the biological activity of the histone methyltransferase (HMT) G9a, a model system.
