In the next-generation sequencing (NGS) workflow, one pipetting or mixing mistake can lead to costly errors. When working with multiple samples, error-free sample handling and proper mixing become even more critical. The Invitrogen Collibri DNA Library Prep Kits prepare libraries for whole-genome sequencing (WGS) on Illumina NGS systems and contain tracking dyes to quickly and easily visualize library prep progress, causing the reaction mixes to change color at each step of the workflow. A homogeneous color of the solution will indicate thorough mixing of the reagents, allowing researchers to have confidence that the process was completed without error. The inert dyes do not interfere with enzymatic reactions and do not compromise sequencing results.

Specifically, the Collibri PS DNA Library Prep Kits for Illumina Systems are suitable for sequencing of genomes of all sizes and variable quality, including FFPE samples. Compared to older library prep kits, the Collibri PS DNA Library Prep Kits provide improved sensitivity of variant detection, which is consistent among low abundance (1 ng) and high abundance (500-1,000 ng) samples. They support PCR-free protocols, helping minimize typical PCR-induced bias. The kits contain adapters with unique dual indexes (UDIs) or combinatorial dual indexes (CDs) that allow pooling of up to 96 different samples prior to the sequencing run. Additionally, the protocol does not require intermediate sample cleanup between End Conversion and Adaptor Ligation steps, which minimizes handling errors and saves time and valuable sample.

Recommended for use for whole-genome sequencing, resequencing and de novo assembly, the kits come with all necessary components, including enzyme mixes, dual-barcoded plate-format adaptors, and cleanup beads, and are applicable for automation of library prep. Not only do the kits make error-free library prep of small sample sizes and low sample quality possible, they also offer higher confidence SNP and INDEL calls resulting from stronger coverage of challenging genomic regions compared to other available library prep technologies.
