Hydrogen bonds between base pairs in nucleic acids can be broken at certain temperatures, allowing double stranded nucleic acids to be separated into single strands. A thermal melt experiment exploits the difference in the number of hydrogen bonds between DNA nucleotides, however the slow temperature ramp rate means that experiments take a long time.

The Agilent Cary 3500 Multizone UV-Vis spectrophotometer accurately controls the temperature of solutions during the experiment.
