Anti-CelTOS Transmission Blocking Activity in vivo and in vitro against P. falciparum by Epitope-specific Monoclonal Antibodies

The presence of CelTOS mAb significantly inhibited oocyst development in mosquitoes in both in vivo and in vitro assays. Importantly, the experimental results with an innovative in vivo humanized mouse model confirmed that circulating anti-CelTOS antibody effectively inhibits P. falciparum ookinete development to oocyst in mosquitoes. These results support the development of CelTOS as a transmission blocking vaccine.

Patients with partial deficiency of recombination-activating genes 1 or 2 (RAG1/2) can present with a wide spectrum of primary immunodeficiencies including combined immunodeficiency with granuloma and/or autoimmunity (CID-G/AI). Prior case reports have highlighted alterations in B and T cell compartments; however comprehensive characterization of these cell populations with focus on autoreactive-prone subsets has not been reported.

Knockout of microRNAs Using the CRISPR-Cas9 System with Paired Synthetic crRNAs

We utilized paired synthetic crRNAs coupled with our synthetic tracrRNA in cells transduced with lentiviral Cas9 to perform a functional knockout on hsa-miR-221. This three-part system (crRNA, tracrRNA and Cas9) has demonstrated efficient gene editing when used with only one guide RNA, but the goal was to use two crRNAs to remove the entire stem-loop.

Gene knockout using CRISPR-Cas9 has dramatically transformed biological research and has been rapidly applied to loss-of-function screening primarily using pooled lentiviral sgRNA libraries. A synthetic CRISPR RNA (crRNA) approach is amenable to screening in arrayed, well-by-well fashion and expands the types of phenotypic readouts that can be used, including high-content and morphology-based assays.

Extracorporeal shockwave therapy accelerates motor axon regeneration despite a phenotypically mismatched environment

A femoral nerve defect model was adapted for the evaluation of proregenerative effects of extracorporeal shockwave therapy (ESWT). Functional evaluation, histology and qRT-PCR data show differences between sensory and motor-derived nerve transplants and a pro-regenerative effect of ESWT. These data provide evidence for the clinical application of ESWT after autologous nerve transplantation as a novel non-invasive method.
