Optimizing mRNA Purification Conditions by Using a High-Throughput Screening Approach

To support the development of mRNA-based therapies scientists require platforms that will help to address the selectivity and capacity requirements for the large-scale manufacturing of mRNA used in vaccine and gene therapy applications.

Typically, mRNA binds to the Oligo dT(25) affinity resin using high ionic strength conditions and neutral pH and is eluted from the column using low ionic strength solutions like water. Although water works well for most mRNA constructs, a need to identify alternative elution buffers which can be used to optimize mRNA recovery, exists.
