This talk will cover two complementary technologies for measuring metabolic flux: Seahorse XF analyzers and liquid chromatography-mass spectrometry (LC/MS) qualitative flux analysis. Both technologies provide important insights into the rates of metabolic reactions within cells, with Seahorse XF measurements providing cellular resolution, and LC/MS qualitative flux measurements providing molecular resolution.

Automated Parallel Derivatization of Metabolites with SWATH-MS Data Acquisition

Suitable quantification standards were generated by derivatized with labelled 13C2-dansyl chloride and 13C2dansylhydrazine. The derivatization of the standards and the urine sample was performed using a PAL RTC autosampler in-line to column-switching LC-HRMS analysis with data independent acquisition (SWATH-MS). The combination of dual DIL with SWATH-MS acquisition enables post-identification of unknown metabolites and quantitation at precursor (MS1) and specific tag fragment (MS2) levels.

We have been using fast scanning TripleTOF systems to analyze a variety of post-translational modifications (PTMs) using both data-independent acquisitions (DIA) and more recently multiple reaction monitoring-high resolution (MRM-HR) to identify, characterize and quantify PTM-containing peptides in a high-throughput format. While rich information has been obtained using traditional CID fragmentation approaches, many of the modifications we study are highly labile and difficult to characterize.
