A new method for generating arrayed RNAi screening tools for any organism
RNA interference (RNAi) using small interfering RNAs (siRNAs) is an important technology for down-regulation of gene expression and a powerful tool to study cellular processes and pathways. Previously, large collections of siRNAs were available only for traditional experimental model systems, such as human and mouse, and predominantly provided as chemically synthesized libraries.

Knockout of microRNAs Using the CRISPR-Cas9 System with Paired Synthetic crRNAs
We utilized paired synthetic crRNAs coupled with our synthetic tracrRNA in cells transduced with lentiviral Cas9 to perform a functional knockout on hsa-miR-221. This three-part system (crRNA, tracrRNA and Cas9) has demonstrated efficient gene editing when used with only one guide RNA, but the goal was to use two crRNAs to remove the entire stem-loop.

Identification of genes involved in cell cycle regulation using arrayed synthetic CRISPR RNA libraries in a multiparameter high-content assay
Gene knockout using CRISPR-Cas9 has dramatically transformed biological research and has been rapidly applied to loss-of-function screening primarily using pooled lentiviral sgRNA libraries. A synthetic CRISPR RNA (crRNA) approach is amenable to screening in arrayed, well-by-well fashion and expands the types of phenotypic readouts that can be used, including high-content and morphology-based assays.

Previous studies suggested that Mycobacterium tuberculosis obtains nitrogen from a diverse range of intracellular nutrients including amino acids. Here, we use a novel system’s based three-pronged approach to define pathways for uptake and assimilation of nitrogen.

Inhibition of The Auto-inflammation Suppressor Protein ISG15 Triggers Preeclampsia by Blocking Trophoblast Migration and Invasion
In summary, ISG15 expression levels are crucial for trophoblast morphology and function (migration/invasion). By blocking trophoblast invasion, reduced ISG15 levels could contribute to impaired spiral artery transformation that reduces utero-placental blood flow in preeclampsia. Thus, agents inducing ISG15 expression are likely to be therapeutic in preeclampsia.

An Emerging Phenotype of Partial RAG 1/2 Deficiency Among Young Children with Autoimmunity and Viral Infections
We describe the natural history of a cohort of 12 patients with confirmed partial RAG1/2 mutations and autoimmunity at a young age. We were seeking the link between viral infections and autoimmunity and tested candidate biomarkers that may reflect the underlying RAG1/2 protein deficiency.
