Integrated DNA Technologies (IDT), has announced the launch of the TriFECTa™ Dicer-Substrate RNAi Kit.
Employing 27-mer duplex RNA, TriFECTa delivers a significant design improvement over first-generation 21-mer based RNAi kits.
Processed by Dicer into 21-mer siRNAs, IDT's 27-mer RNAi design delivers up to a 100-fold increased potency when compared to older 21-mer siRNA models at the same site.
Utilizing the endogenous Dicer system, IDT's TriFECTa represents a substantial improvement over traditional antisense based processes, and gives researchers the confidence they need when studying genes.
Backed by the companies functional knockdown guarantee, at least one of the kit's three Dicer-Substrate RNAi duplexes will deliver a 70% knockdown of the target mRNA when 90% transfection efficiency, and 90% knockdown with positive control is demonstrated.
“Designing RNAi duplexes that engage and exploit Dicer's excellent natural capabilities rather than avoiding it using siRNAs enables researchers to significantly raise efficacy - often at sub-nanomolar concentrations,” noted Dr. Mark Behlke, vice president of molecular genetics at IDT.
“Our research with City of Hope revealed that researchers can achieve dramatically improved levels of RNAi-mediated gene knockdown at significantly lower concentrations than was previously possible.”
The TriFECTa kit includes three target-specific Dicer-Substrate siRNA duplexes (5 nmoles each), Cy3TM DS Transfection Control, fluorescent-labeled transfection control duplex (1 nmole), HPRT-S1 DS Positive Control duplex (1 nmole), DS Scrambled-Neg, universal negative control duplex (1 nmole), RNase Free Duplex Buffer (100mM KAc/30 mM HEPES pH 7.5).
