Proteomic non-small cell lung carcinoma biomarker screening in bronchoalveolar lavage fluid

Using a standardized method to acquire MALDI-TOF proteome profile spectra of bronchoalveolar lavage fluid (BALF), we have shown the upregulation of histatin 3 and calgranulin C in a small pilot cohort of NSCLC patients. This pilot study serves to demonstrate that it is feasible to screen a larger NSCLC patient cohort for BALF proteome level biomarkers in a clinical setting.

A Novel Approach to Internal Standardization in LC/MS/MS Analysis; Sensitive LC/MS/MS Analysis of Gentamicin

A novel approach to internal standardization in LC/MS/MS is demonstrated. Gentamicin, a multi-component aminoglycoside, is derivatized with propyl-d7 chloroformate making the internal standard. Gentamicin components in the samples are derivatized with non-labeled propyl chloroformate making them analogous to the internal standard. Additional benefits of the derivatization are increased hydrophobicity and excellent MS/MS fragmentation.

Predicting hepatotoxicity: Reactive metabolite trapping using glutathione and freshly isolated hepatocytes

This poster presents our results to date using clozapine (a compound known to be associated with GSH-adduct formation) as substrate and using stable isotope GSH (GSH13C2,15N) to enhance specificity. In addition, all analyses have been conducted using an Waters Acquity UPLC-MS/MS. Results we have obtained in hepatocytes are compared against findings using human liver microsomes (HLM).

Single stage reversed-phase liquid chromatography-mass spectrometry for the characterization of triglyceride positional isomers.

The high level of selectivity afforded by RP-HPLC in combination with CID allowed recognition of eight groups of TAG positional isomers. Development of UPLC approach has reduced the run time by half while maintaining separation. The improved method combined with principal components analysis was used to assess the extent of variation of major TAGs in subcutaneous and intermuscular pork fat from four different locations of adipose tissues.

Five noncovalent peptidic ligands show different affinity rankings in solution and gas phase

Stability of noncovalent complexes of VEGF protein with 5 peptidic ligands is studied. Experiments were conducted in solution (NMR CSP, ITC) and in gas phase (CID TOF MS). Each ligand differs from others in chirality of one amino acid. It was shown, that trend of stability of the studied noncovalent complexes is reversed in the gas phase relatively to the solution. An explanation of this behavior is presented.

A library of oxazolidin-2-ones was prepared by solid phase synthesis. Crutial reaction steps were optimized. Highly divers building blocks were selected for the synthesis. All compounds were purified using preparative LC/MS with mass directed fraction collection.

We used Ciphergen technology based on SELDI TOF-MS (Surface Enhanced Laser Desorption/Ionisation Time-of-Flight Mass Spectrometry) for analysis of protein expression profiles of 105 breast cancer tissue samples. The aim of this study was the identification of single proteins or protein patterns specific for different tumour subgroups, which should take advantage of the biomarker as an alternative to commonly used diagnostic and prognostic characteristics.
