AnaSpec has introduced a line of b-amyloid peptides labeled with biotin (N or C-terminal biotin labels) as well as N-terminal biotin labeled b-amyloid peptides with a fluorescein label on the C-terminus.
These peptides have been used in different applications. Nelson, TJ and Alkon, DL. used biotin-LC-b-amyloid-(1–42) in Ab-proteins binding studies. Ab-proteins complex were precipitated with avidin-agarose and western blots run. A paper by Bohrmann, B. et al. reported the use of biotin-Ab(1-40)0) or biotin-Ab (1-42) for their Ab polymerization studies.
Biotin-Ab (1-40) or biotin-Ab (1-42) peptides were incubated in Ab (1-40) or Ab (1-42) coated plates, and streptavidin-peroxidase added. Color development was done using tetramethyl-benzidin, a chromogenic substrate for peroxidase. In the same paper,2 biotin-Ab ( (1-40) incorporation into preformed Ab (1-42) fibrils was reported. Biotin-Ab (1-40) was added to a low concentration of Ab (1-42) fibrils and stained with anti-biotin Ig labeled with colloidal gold and negatively stained with uranyl acetate. A paper by Liu, Y. et al shows the use of biotinylated fibrillar Ab (1-42) in electron microscopy.
Using what they called “FAbB,” fluorescein beta-amyloid (1-40)-Lys(LC-biotin), Leissring, MA. et al. performed proteolysis measurement using fluorescence polarization (FP) technique.
