Microtubules (MTs) are one of the major components of the cytoskeleton, and are composed of dimers of α- and β-tubulin. Microtubule assembly is a dynamic process, and MTs participate in many cellular processes such as structural support and intracellular trafficking.
In neurons, MTs participate in the axonal transport, trafficking of cargo within different compartments of a neuron. MTs are also involved in neuronal cell polarization, neurite branching, and dendritic spine remodeling. Class III β-tubulin is exclusively expressed in neurons and is commonly used as a neuronal cell marker.
Tubulins are subject to post-translational modifications (PTMs) such as detyrosination, glutamylation and acetylation. Some PTMs such as detyrosination reduce microtubule depolymerization, therefore, leading to MT stability. Alterations in MT dynamics and stability is a common feature of many neurodegenerative disorders, and is associated with the ‘dying-back’ of axons.
