Shimadzu Corporation has launched a new improved Accurate Glycan Analyzer(AGA2): a combination of biologically-relevant glycan database and the unique MSn properties of the AXIMA® Resonance.

Glycans play an important role in biological processes and are involved in protein interactions as well as cell surface recognition mechanisms. Despite this they have received little attention.

This is in part due to the complex branched structure of the compounds which has meant that their study and structural identification has been a highly specialized discipline: Until now!

Combining the simplicity of MALDI, the power of MSn, and the accuracy and resolution of Time-Of Flight, the AXIMA® Resonance provides a unique solution for the next generation of complex structural and sequencing challenges.

The AXIMA Resonance features high mass resolution and mass accuracy across MS and MSn analyses. This ability to carry out simple high-resolution precursor ion selection for MSn experiments makes it ideal for the study of glycans.

The AXIMA Resonance is coupled to a database containing glycan structural information generated from actual MSn spectra of to select precursor peaks up to MS4.

The resulting spectra are interpreted to return the most likely structure. The database has been created from glycans synthesized by the National Institute of Advanced Industrial Science and Technology (NJAIST) in Tsukuba, Japan, and contains only well-defined and characterized, biologically relevant, glycans.

As well as adding new species to the database, the AGA2 system now allows for the use of common fluorescence labeling schemes including 2-aminopyridine (2-AP / PA), 2-aminobenzoic acid (2-AA) and aminobenzamide (2-AB), or without labelling.
