Octet® Label-Free BLI Detection Systems

Octet® Bio-Layer Interferometry (BLI) Detection Systems provides research labs with a versatile label-free interaction analysis solution with a diverse range of applications from determining biomolecular binding affinity and kinetics to concentration analysis of proteins and antibodies.

Key Octet features that enable scientists to accelerate experimental timelines include:

  1. Microfluidics-free sample delivery allows for faster time to results, low instrument maintenance, and low operational expenditures
  2. A wide range of available biosensors to choose from contributes to fast and flexible assay development
  3. Crude sample compatibility allows for minimal sample preparation and early screening of analytes from unpurified matrices
  4. Intuitive, easy-to-use data analysis software tools

Since the COVID-19 genomic sequence was first shared in January of 2020, Octet data have been featured in many breakthrough publications dissecting COVID-19 disease mechanisms, vaccine mechanism of action studies, therapeutics development workflows, and serological assays to facilitate COVID-19 antibody testing. Key Octet applications areas include:

Kinetic and affinity analysis

The Octet family of instruments provides researchers with a convenient tool for rapid kinetic, affinity, and stability analysis of vaccine candidates and different vaccine types, including whole inactivated viruses, virus-like particles, and recombinant proteins during the development cycle. Kinetic and affinity analysis are also essential in therapeutics development workflows, whether it is a small molecule, peptide, or antibody development program. Lack of microfluidics in Octet systems enables early screening of lead candidates in crude matrices reducing extra purification steps.

Titer and concentration analysis

Octet platform’s simple biosensor dip and read format and the use of easy-to-customize disposable biosensor surfaces enable researchers to screen for clones during cell line development, measure antibody and virus titers (e.g., AAV, influenza, etc.), detect and monitor process generated contaminants such as host cell proteins and residual protein A with better precision and robustness than traditional ELISAs.

Epitope binning analysis

Epitope binning helps identify antibodies that block the same epitope on a target antigen and is crucial for identifying or engineering mAbs with favorable target recognition profiles. Octet's high-throughput capabilities, availability of various biosensor chemistries, and the ability to characterize antibodies directly from hybridoma supernatants or lysates make the Octet the easier and faster choice for epitope binning assays.
