Application Notes
Contaminant bovine IgG assay
Application NoteMolecular Devices
Fetal calf serum containing bovine IgG is commonly used in mammalian cell culture media for the production of recombinant pharmaceuticals. During the purification process of a recombinant pharmaceutical, bovine IgG may be copurified with the product, and may represent a biological hazard for clinical use. Therefore, it may be necessary to assay for contaminant bovine IgG in a final product originating from mammalian cells.
READ MOREApplications of rapid immunoassays (FAST ILA) in cell culture/fermentation and process development using the Threshold System
Application NoteMolecular Devices
Accurate measurement of the concentration of a biological product is important in bioreactor/fermenter monitoring, downstream processing and final product quality control. Currently, electrophoretic, chromatographic and immunochemical methods are the most commonly used. These three methods are currently used in combination to provide the desired sample information.

Application NoteMolecular Devices
Reporter-gene assays are used to study eukaryotic gene expression. Dual genetic reporters are commonly used in transient transfections of cultured cells to minimize experimental variability. Recently, Promega introduced a Dual-Glo Lucoferase assay system which replaced the outdated ‘flash’ assays used previously. Ensuring both genetic reporters have different substrate requirements enables them to be detected using a single reaction mixture.

Application NoteMolecular Devices
The proteasome is a massive protein complex which can recognize and bind targeted proteins, and inject them into the central core where the proteins are successively degraded into short peptides. Whereas protein synthesis is well-understood after decades of study, major advances in our knowledge of protein degradation have occurred only in the last two decades.

Application NoteMolecular Devices
Calcium release-activated calcium (CRAC) channels play an important role in intracellular Ca2+ homeostasis. Like other store-operated calcium channels, currents through the CRAC channel (ICRAC) are activated by depletion of calcium in the endoplasmic reticulum (ER) and serve the purpose of slowly replenishing the ER with Ca2+.

Application NoteMolecular Devices
The Threshold® Immuno-Ligand Assay (ILA) adapts reagents used in ELISA, RIA or radioreceptor assays for the Threshold System by covalently attaching biotin or fluorescein labels to the binding proteins and/or ligands. To simplify labeling and purification of proteins, Molecular Devices has formulated biotin and fluorescein as N-hydroxysuccinimide esters.

Application NoteBMG Labtech
The BlueScreen HC assay from Gentronix utilizes a luminescence-based reporter system to measure genotoxicity. The assay generates positive results e.g for direct-acting mutagens and clastogens and correct negative results for non-carcinogens. In addition, reduced cell proliferation, a measure of cytotoxicity, is assessed, too. Using the FLUOstar Omega microplate reader from BMG LABTECH the BlueScreen HC assay has been shown to be fast, accurate and reproducible.

Application NoteBMG Labtech
Campylobacter are Gram negative microaerophilic organisms which are a common commensal of mammals and birds and are one of the most frequently isolated causative agents of human bacterial enteritis worldwide. Infection in humans is frequently associated with the consumption of under cooked poultry and is generally self limiting though it can in a small number of cases result in severe complications such as Guillain-Barre syndrome.
