A number of GPCRs activated by peptide ligands were assessed using different assay technologies. It was observed that the Ec50 values obtained using htrf assays were significantly higher than those obtained with other assay platforms. This difference was not observed with a non-peptide liganded GPCR. This highlights that with the GPCRs tested the use of htrf technology may not be the most appropriate assay format for peptide liganded GPCRs.
We utilized paired synthetic crRNAs coupled with our synthetic tracrRNA in cells transduced with lentiviral Cas9 to perform a functional knockout on hsa-miR-221. This three-part system (crRNA, tracrRNA and Cas9) has demonstrated efficient gene editing when used with only one guide RNA, but the goal was to use two crRNAs to remove the entire stem-loop.READ MORE
